Gene Expression CoverageAs of AgamP3.6 the Affymetrix microarray platform contains Anopheles probes corresponding to 11,033 annotated protein-coding genes, equating to 82% of the genes in the genome. After normalization and quality control for MozAtlas v1.5 (21/7/2011), we detect expression in 9,831 probes corresponding to 7388 unique genes across Anopheles tissues and in whole body samples.Approximately 24% of Anopheles transcripts in dissected samples are absent from whole-body estimates. |
Gender-Biased ExpressionA linear model was fit to male and female tissue samples. On the basis of differential expression, probes are classified as either male-biased or female-biased with a 2-fold change of intensity and statistical significance at the Q<0.05 level. Overall, 56% of genes are sexually dimorphic in at least one organ. Of the 4138 sexually dimorphic genes, 70% are detected in whole-body male and female samples, with the remaining 30% only in dissected tissues. The proportion of sexually dimorphic expression in each tissue versus total sexual dimorphism is provided below, including the ratio of female- to male-biased expression (female:male). Chromosome Distribution of Germline and Somatic genesTestis and Ovary expression sets were derived from genes detected in either the testis or ovary. Genes were considered to have acquired sex-biased somatic expression if one or more tissues show elevated expression, using calls of differential expression between males and female where there was a significant 2-fold change of expression intensity in one sex. This analysis provided gene sets for analysing expected versus observed chromosome distributions amongst the autosomes and X chromosome via chi-squared tests. A Fisher's exact test was further used to determine over- or under-representation of single chromosomes against expected counts (highlighted in bold).
